Reactive oxygen species (ROS) induce the oxidative stress, programmed cell death and the expression of many cytokines. p-Hydroxybenzyl alcohol (HBA) is known to play a pivotal protective role against inflammatory and oxidative stress-related diseases. We have developed a new biodegradable multifunctional peroxalate copolymer containg HBA (HPOX). Peroxalate ester linkages in the backbone reacted with and scavenged hydrogen peroxide, leading the release of HBA. We found that HPOX nanoparticles exerted stronger antioxidant, anti-inflammatory and anti-apoptotic activities than free HBA in antioxidant activities. HPOX nanoparticles effectively reduced generation of nitric oxide, tumor necrosis factor-alpha (TNF-a) and suppressed apoptisis of stimulated RAW 264.7 cells. Therefore, we suggest that HPOX nanoparticles are highly potent for the treatment of oxidative damage-related diseases. |
