Injectable drug delivery systems using thermosensitive hydrogels offer several advantages such as no surgical procedure for local drug delivery and simple formulation with therapeutic drugs. Novel biodegradable and thermosensitive polyphosphazene-paclitaxel conjugates bearing hydrophobic L-isoleucine ethyl ester, hydrophilic α-amino-ω-methoxy-poly(ethylene glycol), and paclitaxel have been synthesized for local delivery of anticancer drug. The aqueous solution of conjugates showed solution state during injection and formed a gel at injection site. The conjugate gels showed good in vitro antitumor activity against several tumor cell lines evaluated by MTT assay. Also, the conjugate gels exhibited effective inhibition of tumor growth compared to other control groups using human gastric cancer cell line on nude mice. Polyphosphazene-paclitaxel conjugate hydrogel can be a good candidate for treatment of localized solid tumor with a simple injection. |
