Although organ transplantation is regarded as an effective remedy for organ failure, the shortage of donor organ and the immunological rejection have been considered as a challenge in clinical practice. Alternatively, tissue engineering technique which integrates 3D scaffolds, cells, signal molecules is recently introduced and spotlighted in research and clinical fields. However, necrotic center occurred in 3D cell/scaffold caused by insufficient supply of oxygen into the scaffold is remained a huge huddle for producing a clinically relevant volume of tissue/organ. Cell behaviors (proliferation, apoptosis, protein synthesis, etc.) in hypoxia were evaluated for 28 days. And osteogenic differentiation of cell (ALP activity, calcium deposition, immunocytochemistry, and RT-PCR) after 10 days exposure in hypoxic condition was also investigated. New bone formation (histology, CT) of the cell-loaded PFO-HP and PBS-HP was also investigated using a miniature pig (mandibular defect model). |
