Chemically-Conjugated Bone Morphogenetic Protein-2 on porous PCL/BCP-Collagen scaffolds stimulates Osteogenic activity |

The conjugation of recombinant human bone morphologenetic protein-2(rh-BMP-2) on absorbable porous PCL/BCP-Collagen scaffold was developed in this study. Immunofluorescent staining against BMP-2 demonstrated that BMP-2 was successfully immobilized on the PCL/BCP-Collagen three-dimensional scaffold by aminolysis and subsequent chemical conjugation. The ability of this system to deliver rhBMP-2 was tested for the repair of bone defect. The morphology and pore size distribution of PCL/BCP-Collagen and PCL/BCP-Collagen-BMP-2 was observed by Scanning Electron Microscope (SEM) and Mercury porosimetry. The in vitro evaluation of with and without rh-BMP-2 scaffolds was done by MTT assay, Live/dead cell and immunofluorescent with mouse calvaria-derived preosteoblastic cells (MC3T3-E1). The surface modification of PCL/BCP-Col scaffolds with BMP-2 showed the good ability to support the attachment and proliferation of the pre-osteoblast cells. The PCL-BCP-Collagen-BMP-2 could be used for cell and growth factor carriers for bone regeneration. |
