제목 Evaluation of gene transfection efficiency of stem cells using DNA/polyethyleneimine complexes
초록 The aim of this study was to evaluate transfection efficiency in rat bone marrow stem cells (rBMSCs) and rat muscle-derived stem cells (rMDSCs) using DNA/polyethyleneimine (PEI) complexes, because a few studies have compared the transfection efficiency at both stem cells. In the characterization of PEI/DNA complexes, gel electrophoresis showed that DNA completely binds to PEI at a N/P charge ratio of 1-2. The N/P charge increasing exhibited low cell viability, indicating that increasing amount of PEI affected the cytotoxicity. Transfection with PEI/DNA complexes to rBMSCs with N/P charge ratios of 1-32 gave transfection efficiencies of 1-12%. However, transfections with PEI/DNA complexes to rMDSCs were below 5% at all N/P charge ratios. The transfection efficiency of rBMSCs was higher than that of rMDSCs. In the current study we compared the transfection efficiencies of rBMSCs and rMDSCs using PEI. We found that gene delivery using PEI is relatively inefficient to rMDSCs.
