Polyethyleneimine-Mediated Gene Delivery into rat mesenchymal stem cells |

Recently, a number of new techniques have been developed to deliver DNA into target cells. Among the cationic polymer-based formulations tested for non-viral gene transfections, polyethyleneimine (PEI) is considered one of the most efficient polymers for gene delivery. In this study, we examined the use of PEI as a non-viral gene carrier for rat bone marrow stem cells (rBMSCs) and rat muscle derived stem cells (rMDSCs). The formation of PEI-DNA complexes was characterized by gel electrophoresis and measurement of particle size, surface charge and loss of fluorescence. As the proportion of PEI was increased, the size of the PEI-DNA complexes decreased from 470 to 150 nm, the positive surface charge increased from -25 to 25 mV, and the fluorescence intensity decreased from 45 to 10. The transfection efficiency of rBMSCs was highest at N/P charge ratio 4 (16%) and that of rMDSCs was highest at N/P charge ratio 16 (5%). |
