제목 A ratiometric fluorscent probe for detection of Histone deacetylase activity
초록 Histone deacetylase (HDACs) play important roles in regulating various physiological and pathological processes such as gene expression, cell cycle, and tumorigenesis. These enzymes activity can be attractive targets for drug development. In this study, we synthesized a ratiometric fluorescent probe 1 by incorporating the Kac residue and the S-NBD group. Upon enzymatic removal of the acetyl group in the Kac residue, the released free amine reacted intramolecularly with the S-NBD moiety, resulting in increase the dynamic range of fluorescence measurements and shift the emission spectra. A ratiometric fluorescent probe 1 is superior for allowing the measurement of emission intensities at two different wavelengths, which should provide built-in correction for environmental effects, and they also increase the dynamic range of fluorescence measurements.
