Various chromatography purification processes are used in the manufacture of biopharmaceuticals, and affinity chromatography, in which a target antibody protein is separated using a ligand that acts only on a specific protein, is representative. In particular, ‘Protein A’ has a selective binding force to the antibody, so it is possible to increase the efficiency of drug production and reduce costs. In order to increase the number of reuse of the resin, cleaning-in-place (CIP) should be performed to remove various contaminants remaining after antibody purification. However, resin for protein-based purification has weak alkali tolerance, so it is necessary to develop stable immunoglobulin-binding proteins under alkaline conditions. We have developed Protein A chromatography resin for antibody purification by developing improved alkali tolerance and agarose bead manufacturing technology. |
