Utilization of abundant and cheap carbon sources can effectively reduce the production cost and enhance economic feasibility. Acetate is a promising carbon source to achieve cost-effective microbial processes. In this study, we demonstrated the efficient conversion of acetate into itaconate. The metabolic pathway was initially reconstructed by amplification of linear pathway and optimization of the glyoxylate shunt pathway. The resulting strain showed significantly enhanced itaconate production by 6.5-fold also with enhanced acetate assimilation. However, the titer and yield could not exceed those from glucose and there is still room for enhancement. Therefore, evolutionary engineering was additionally applied with novel screening device. Adaptive evolutionary engineering (ALE) could enhance the titer and yield by 2.5-fold and 7.2-fold, respectively. The Whole Genome Sequencing (WGS) will be conducted and the effective mutant will be validated for construction of optimal producer. |
