Hypoxia-Sensitive Self-Immolative Dendrimer for Fluorescence Imaging |

Self-immolative dendrimer can undergo triggered degradation by modified trigger which attached dendrimer core. End group of conjugated molecules undergo domino-like degradation by single triggered event of self-immolative dendrimer. Also, dendrimer has well defined 3-D structure and smaller size than same molecular weight of polymer, provide easier access to limited area like cancer solid tumor. In this study, we modified hypoxia sensitive trigger to apply diagnose hypoxic cancer. Most solid tumor has hypoxic condition because it located far from blood vessels, which provides the conditions of low oxygen level and abnormal tumor tissue environment. Nitro group for hypoxia-sensitive trigger at dendrimer core and fluorescent dye was modified at dendrimer tail end for drug delivery application. Nitro group undergo reduced by metabolic reduction at hypoxic condition, cascade reaction progressed by carbamate linkage and release anticancer drug which conjugated at the periphery of dendrimers |
