제목 | Pharmaceutical preparation of BCNU for increasing the stability to hydrolysis and release behaviour of self-microemulsified BCNU from PLGA wafer |
초록 | 1,3-bis(2-chloroethyl)-1-nitrosourea(BCNU) is an effective chemotherapeutic agent which has been used clinically for treating malignant brain tumor. But it is unstable to heat and light, and also has a half life of 20min in vitro and less than 15min in vivo by hydrolysis. To improve the stability of BCNU to hydrolysis in body fluid and blood, self-microemulsion system including oil, contrast medium, and contrast agent was prepared. Biodegradable polymeric implant of the self-microemulsified BCNU was prepared using PLGA as the biodegradable matrix. Self-microemulsion system was established for enhancement of BCNU stability. Self-microemulsified BCNU resulted in a significant 2.9-fold increase of half-life in vitro . Release of self-microemulsified BCNU from PLGA wafer followed 1st order release kinetics. The emulsion particle size was determined by the dynamic light scattering and degradation behavior of wafer during the release test was evaluated by scanning electron microscopy (SEM).
