Tyrosinase is an essential enzyme that mediates melanin synthesis in melanocytes to protect skin from UV radiation and radical species. Skin diseases such as albinism and vitiligo can occur if melanocytes stop producing functional tyrosinases. One promising way to treat these skin diseases is delivering tyrosinase directly to the skin to generate synthetic melanin, but the skin barrier is a considerable bottleneck for protein delivery. Therefore, we designed a carbon nanomaterial-based nanocarrier by a surface modification to be compatible with skin tissue. We analyzed the characteristic properties and dimensions of the tyrosinase-immobilized nanocarriers for topical skin delivery while maintaining tyrosinase activity. Also, we demonstrated that tyrosinase was efficiently delivered into ex vivo porcine skin by confocal Raman microscopy. This study shows a model system for topical skin protein delivery using nanocarriers. |
