Biosynthesis and Characterization of Growth Factor Receptor Binding Polypeptides for Advanced Drug Delivery |

A specific ligand binds to a growth factor receptor (GFR) in the cell membrane, which could trigger a wide variety of intracellular signaling. In this study, GFR binding fusion polypeptides composed of thermally responsive elastin-based polypeptides (EBPs) and GFR targeting peptides were genetically engineered, expressed, purified and their physicochemical properties were characterized. The EBPs were introduced as non-chromatographic purification tags while the GFR targeting peptides being presented for GFR targeted drug delivery. Especially, introduction of a series of EBP diblock copolymers to the GFR targeting peptides resulted in formation of temperature-triggered core-shell micellar structures with multivalent GFR binding peptides under physiological conditions, which could largely enhance their binding affinity to GFRs. This work suggests that the thermally responsive GFR binding polypeptides would open a new avenue for advanced drug delivery system. |
