제목 Photothermal mediated gene delivery using IR780 loaded bovine serum albumin coated cell penetrating peptide conjugated disulfide crosslinked polyethylenimine polyplex in breast cancer cells.
초록 Photo-triggered gene release can play a pivotal role in minimizing these limitations and achieve high gene transfection efficiency with minimum toxicity. In the current study, a photothermally controlled gene releasing nanocomplex was developed by complexing peptide conjugated ssPEI polyplex with IR780 loaded bovine serum albumin (BISPD). This BISPD nanocomplex forms a stable nano-sized complex with plasmid DNA and shows higher gene transfection efficiency without any sign of cell toxicity in 4T1 and 4T7 breast cancer cells. Moreover, the BISPD nanocomplex demonstrates an enhanced gene transfection efficiency upon NIR irradiation from 808 nm laser, which is attributed to accelerated endosomal escape of polyplexes augmented by locally induced heat in endosomal vesicles. To investigate the endosomal escaping effect of the BISPD nanocomplex, Bafilomycin A1 was employed and it confirmed the endosomal escape was mediated by the local heat generated by BISPD complex upon irradiation.
