GENTICALLY MODIFIED EXOSOMES AS DELIVERY VEHICLES OF SMALL INTERFERING RNA FOR STEM CELL ENGINEERING AND IN VIVO DELIVERY |

The present study explores the versatility of bioengineered exosomes as potent siRNA carriers for stem cell engineering and in vivo delivery. Exosomes were engineered by overexpressing exosomal membrane proteins fused with cell adhesion or cell penetrating peptides to enhance intracellular gene transfer. The bioengineered exosomes mediated highly effective siRNA delivery with negligible cytotoxicity and resulted in enhanced lineage specification of human adipose-derived stem cells and human neural stem cells. Furthermore, we found that that engineered exosomes internalize into target cells through clathrin-mediated endocytosis and micropinocytosis. Finally, we show that exosomes are also efficient in delivering siRNA in vivo with no detectable hepatotoxicity.This work was supported by a grant (2015R1A2A1A15053771) from the National Research Foundation of Korea (NRF). |
