제목 Programmable construction of a DNA origami Nanostructure formed by selective 5’ cohesive ends ligation based on Thermodynamic and Stereochemistry
초록 As a breakthrough for usage of conventional genetic materials, DNA creatures have been developed. Two different methodologies have been proposed for new DNA architectures. They represent (1) One is DNA scaffold method which uses a long single-strand as a template while small DNA fragments as staple DNAs are used and (2) the other is enzyme-catalyzed brick-and-mortar method which links small DNA nano-blocks via liagse activity process. Although the former has been generally used to fabricate large DNA constructs, the latter has shown a merit to easily make complex DNA structure in small dimension. Here, we improved in such ligation. In consideration of sequence arrangement on overhang sequences, both purity and yield of anisotropic DNA product were significantly influenced. Inspired by these achievements and as engaged with fluorescence dye, such DNA construct was utilized for use of effective detention and real-time diagnosis of cancer-specific RNA markers.
