제목 Bone regeneration with neuropeptide substance P coupled with self-assembling peptide nanofiber in a rat calvarial defect model.
초록 In this study, bioactive KLD12(KLD12+KLD12-Substance P) was developed to mimic endogenous tissue healing process. PLA(Poly lactic acid)/β-TCP(beta-tricalcium phosphate)+bioactive KLD12 was applied to repair calvarial defect. The structures of KLD12, KLD12-SP, bioactive KLD12 were observed by TEM and CD spectra. Bioactive KLD12 nanofibers(5-10nm) were created in physiological conditions by forming β–sheet structure. In vivo MSCs recruiting ability of bioactive KLD12 was observed by using in vivo fluorescence imaging system I-VIS. It was demonstrated that the i.v. injected NIR-labeled hBMSCs were recruited to the scaffold containing bioactive KLD12. Bone tissue regeneration was evaluated by observing gross morphologies H&E and MT staining datas of 12w and 24w after surgery. From the results, it was considered that PLA/β-TCP+bioactive KLD12 composite construct enhance bone tissue regeneration without cell transplantation.
