Recently, the incorporation of stimuli-responsiveness on the surface of mesoporous silica nanoparticles (Si-MP) has allowed for the exploitation of new drug delivery vehicles. Si-MPs have unique features such as stability, biocompatibility, controllable pore size, and readily modified surface functionalities. The large surface area and pore volume of Si-MP have offered the possibility of encapsulating and delivering large quantities of drug molecules. In this study, we report on the novel Si-MP covered with pH-controlled bioabsorbable CaP nano-coatings that allow the facilitated release of entrapped drug molecules by responding to intracellular endosomal pH. The key idea is the urease functionalization of Si-MP surfaces and the subsequent enzyme-mediated surfaced mineralization. The CaP coating could hold the encapsulated drugs in extracellular condition, whereas it could be dissolved within intracellular endosomes to accelerate the drug release. |
