Thermoresponsive fiber-based microwell scaffolds capable of formation and retrieval of salivary gland stem cell spheroids for the regeneration of damaged salivary glands |
초록 |
3D spheroid culture enhances cell-cell interactions and promotes the expression of stem cell properties, yet application of the spheroids is challenging. We fabricated a thermoresponsive fiber-based scaffold for formation and retrieval of spheroids with enhanced expression of stem cell-related genes than salivary gland stem cells in a monolayer culture(SGSCmonolayer). SGSC-derived spheroids(SGSCspheroid) were then implanted into the submandibular glands of mice after fractionated X-ray irradiation. The enhanced production of submandibular acini specific mucin and preservation of MIST1+ mature acinar cells were observed in SGSCspheroid group. SGSCspheroid group exhibited greater amelioration of tissue damage, preservation of KRT7+ terminally differentiated luminal ductal cells and less cellular damage than SGSCmonolayer group. A significant increase in Ki67+ proliferative acinar cells were noted only in SGSCspheroid group. Our results suggest that this scaffold facilitates the production, retrieval and delivery of SGSCspheroids to damaged salivary glands for therapeutic purpose. |
