제목 Reactive Oxygen Species Responsive Pheophorbide A-Glycol chitosan Nanoparticles for Photodynamic Cancer Therapy
초록 We have designed amphiphilic reactive oxygen species (ROS) responsive nano-carrier for photodynamic therapy (PDT) by conjugating glycol chitosan and pheophorbide A with ROS responsive thioketal linker (GC-TK-PheoA). In aqueous solution, the amphiphilic GC-TK-PheoA bioconjugates self-aggregated to form core-shell structured nanoparticles through the hydrophobic interaction of PheoA. The resulting GC-TK-PheoA nanoparticles possess good colloidal stability. The photoactivity of GC-TK-PheoA nanoparticles was greatly suppressed in an aqueous environment by the self-quenching effect, which enabled the GC-TK-PheoA nanoparticles to remain photo-inactive and in a quenched state. However, the nanoparticular structure instantaneously dissociated in the presence of ROS, causing an efficient dequenching process. These results suggest that GC-TK-PheoA nanoparticles have good potential as a smart PDT nanoparticular system for anti-cancer therapy.
