Cells control gene expression by genetic circuits for interactions with environments and specific performance. Designing a synthetic genetic circuit becomes essential because of its diverse application. But understanding the interactions of biological components is crucial for efficient and accurate design. In this study, we standardized and evaluated the transcription factor-based genetic logic gates to describe their performance and relationships. We depicted a mathematical response function based on the Hill equation. The input and output were normalized with the standard promoter activity unit. We achieved 12 NOT gate response functions. Furthermore,we analyzed the change of response function depending on the component variation. We can apply this method to other genetic modules. In conclusion, we characterized transcription factor-based genetic logic gates so that we can describe the relationships of genetic circuit components. |
