Bacterial infection is a major cause of peritonitis that leads to severe sepsis. The treatment of the peritonitis depends on achieving therapeutic concentrations of antibiotics within the body fluids and target tissues. Local delivery of antibiotics offers several advantages, including fewer side effects and higher local concentration of a given medicine compare to systemic one. However, the burst release of commonly used antibiotics due to their water-soluble property has been considered as a practical limitation in local delivery. In this study, we prepared ion-complexed tetracycline-loaded alginate sheets which can allow sustain release of antibiotics and firmly protect a defect of gastrointestinal tract, and thus can be an ideal system for the treatment of peritonitis. Their drug release behavior, cytotoxicity, and anti-bacterial effect were investigated. In vivo animal study to evaluate their remedial value for the peritonitis was also conducted using a rat model (cecum defect). |
