Smart polymeric carriers recently gained much attention in anticancer therapy to enhance the efficient drug release in tumor tissues. We prepared a series of triblock poly(N-isopropylacrylamide)75-block-poly(L-Lysine)35-block-poly(L-Histidine)n (p(NIPAM)75-b-p(Lys)35-b-p(His)n) (n=35, 50, 75, 100) polymer bio-conjugates through combination of reversible addition fragmentation chain transfer (RAFT) and ring opening polymerization (ROP) of α-amino acid N-carboxyanhydrides. The dual stimuli-responsive nature of the triblock polymer is confirmed by nuclear magnetic resonance and dynamic light scattering. Moreover, in vitro drug release profile demonstrated that the maximum release of Dox shows in pH 5.0 at 37 oC rather than pH 7.4, and pH 6.0 at 37 oC. The results reveals that these dual stimuli responsive hybrid system expecting to be nice choice for the further studies in anticancer therapy. |
