Minoxidil is one of the most effective drugs for the treatment of hair loss. However, because minoxidil has a poor solubility in water, its application has been limited due to the toxicity of solvent and the low delivery efficiency into skin tissues. In this work, we developed a water soluble hyaluronate -minoxidil conjugate (HA-minoxidil) for transdermal treatment of hair loss. HA-minoxidil was synthesized by BOP chemistry and its successful synthesis was confirmed by HPLC and NMR analysis. HA-minoxidil formed a spherical nanoparticle by hydrophobic interaction of minoxidil, which can be used to encapsulate other hydrophobic drugs for a combination therapy. The formation of HA-minoxidil nanoparticle was confirmed by DLS and TEM. We will present intravital two-photon microscopy to visualize the transdermal delivery of HA-minoxidil and in vivo evaluation of hair regeneration in hair loss model animals of Albino Webster mice and Mdx mice. |
