제목 Hyaluronic Acid - Epidermal Growth Factor Conjugates for Transdermal Wound Healing
초록 Epidermal growth factor (EGF) has been known for its excellent therapeutic effect on wound healing. In this work, EGF was conjugated with hyaluronate (HA) and developed into patch-type wound healing agent. HA-EGF conjugate has been synthesized by coupling reaction between aldehyde-modified HA and N-terminal amine groups of EGF, and the synthesis was confirmed by GPC analysis. Biological activity of EGF after HA conjugation has been demonstrated using ELISA and proliferation test of fibroblasts. To facilitate transdermal delivery of HA-EGF conjugate, the conjugate was incorporated in the polymeric patch composed of physically-crosslinked HA. Two-photon microscopy visualized more efficient transdermal delivery of HA-EGF conjugates to both normal skin and peripheral tissues of the wound site rather than EGF. Furthermore, in vivo transdermal delivery of patch-type HA-EGF conjugate resulted in significantly improved regeneration of skin tissues even into hyperdermis.
