The injection of bulking agents has usually been used as the first line therapy for the treatment of urinary incontinence. However, the injection volume decrements with time caused by particle migration and resorption in the body have been considered as a critical problem. In this study, we prepared pDNA (encoding for bFGF)/PEI-PEG complex-loaded polycaprolactone (PCL) particle/poly(DL-lactic-co- glycolic acid) (PLGA) mixture as an injectable bioactive bulking agent which can provide bulking effect and stimulate the defect tissues around urethra for the effective treatment of urinary incontinence. The release behaviour of the pDNA/PEI-PEG complex from the mixture and the bFGF expression from the gene transfected cells were investigated. The mixtures were also subcutaneously applied into the back of hairless mouse to evaluate their initial bulking effect, biocompatibility and tissue responses (Grant sponsor: NRF-2011-0001804). |
