Efficient delivery of antigen to the antigen presenting cells (APCs) and their subsequent antigen presentation to initiate the production of activated T cells is vital parameter in cancer immunotherapy. However, poor uptake efficiency of APCs limits the production of sufficient activated T cells. For the effective delivery and activation of APCs, we have developed dextran functionalized reduced graphene oxide (rGO-dextran). Graphene have high surface area, hydrophilic and hydrophobic surface that can be used for the high loading of ovalbumin (OVA). Dextran on the graphene surface provided good colloidal stability by exposure of their large amounts of hydroxyl groups and also facilitate cellular uptake by interaction with carbohydrate receptor present on DCs. The efficient OVA delivery to the DCs using rGO-dextran was confirmed by FACS. Activation of DCs and T cells was confirmed from analyzing the expression of specific surface marker on cell surface. Finally, prophylactic vaccination study shows that OVA-loaded rGO-dextran inhibited the tumor growth after vaccination. The proposed rGO-dextran materials can be used as vaccine delivery application in cancer immunotherapy. |
