Mussel Protein-Based Injectable Sticky Microspheres for Stem Cell Delivery |

Microsphere (MS) are attractive as injectable cell carriers for stem cell therapy, because they can be accurately adapted to tissue defects through minimally invasive surgical procedures, while allowing efficient cell loading. We present a sticky MS using a recombinant mussel adhesive protein (MAP) fused with a vascular endothelial growth factor (VEGF)-mimicking peptide. The MSs were uniformly constructed by oxidation-induced covalent crosslinking of 3,4-dihydroxy-L-phenylalanine (DOPA)-containing recombinant MAP-VEGF within a microfluidic device. The fabricated MAP-VEGF-MSs exhibited excellent elasticity for retaining their shape and integrity during a syringe-injection. It was demonstrated that the MAP-VEGF-MSs can provide an efficient stem cell loading, support cell spreading as well as enhance cell proliferation. In addition, the cell-laden MAP-VEGF-MSs showed the potential to directly and actively participate in osteogenesis and vasculogenesis in vitro and in vivo, in a minimally invasive, site-directed manner. |
