Non-viral vectors contineue to attract a great deal of interest due to advantageous safety profile. Among the non-viral vectors, Chitosan is considered to be a good candidate for the gene delivery system, since cationically charged chitosan can be complexed with negatively charged plasmid DNA. Chitosan is the second most plentiful biomass and is already known as a biocompatible, biodegradable, and low-toxic material. In this report, low molecular, water-soluble chitosan (LMWSC, molecular weight of 22kDa) was characterized and evaluated as a gene carrier. The formation of plasmid/LMWSC complex was proved and transfection efficiency in 293T cells was evaluated. The cytotoxicity of LMWSC also was determined by MTT assay. This study showed that LMWSC is one of the candidates for DNA deivery. And artery wall cells targeted gene carrier was synthesized to therapy the disease related with cardiovascular, such as atherosclerosis, restenosis using LMWSC. AWBP-PEG-g-LMWSC was synthesized by conjugation the artery wall binding peptide (AWBP), a specific targeting peptide, to the end of PEG-g-LMWSC as a gene carrier to target artery wall cells. Characters of the synthesized AWBP-PEG-g-LMWSC were analyzed by measuring FT-IR, 1H-NMR, zeta-potentiometer, and atomic force microscopy (AFM). The data from this study provided useful and important information for the future application of in vitro, in vivo studies.
