Mesenchymal stem cells (MSCs), which is multipotent stromal cells, has been clinically administrated for graft-versus-host disease, sepsis, paralysis and stroke by direct intravenous injection. In order to increase cellular stability during administration, we have prepared multilayer films using RGD peptide on individual MSCs, resulting that film coated MSCs showed significantly increase cell survival during agitation culture which is mimicked blood vessel, having cytoprotective effect of film which could be also supported by the activation of survival-related protein, Akt. The multilayer film does not interfere cell differentiation, attachment and migration, keeping that cellular openness after coating. In addition, the multilayer films showed the increased survival rate in blood circulation and stem cell recruitment at injured area. Therefore, in this study, we concluded that the RGD-peptide film is powerful tool for in vivo transplantation in stem cell therapy. |
