제목 CD44-targetable chemotherapy of human gastric cancer cells  using pH-responsive π-hyaluroanan nanocarrier
초록 In this study, we demonstrated targeted delivery potential and therapeutic effect of doxorubicin-loaded pyrenyl hyaluroanan nanocarrier (HPDOX) to CD44-overexpressed gastric cancer cells (MKN-45) and their in-vivo model. We synthesized pyrenyl hyaluroanan composed of hydrophilic backbone of hyaluroanan which is substrate of CD44 and pyrenyl conjugates providing π-rich region for loading doxorubicin. DLS, AFM, and TEM studies confirmed that HPDOX were successfully prepared. Release profiles of HPDOX depending on pH showed that release of doxorubicin was triggered at tumor pH condition by breaking of π-π interaction between pyrenyl group and protonated doxorubicin. CD44 targetability and therapeutic effect of HPDOX in MKN-45 cell line were verified with confocal fluorescence microscope study and MTT assay, respectively. Finally administration of HPDOX in MKN-45 xenograft animal model effectively suppressed growth rate of tumor compared with free doxorubicin administration.
