Glioblastoma multiforme is most aggressive malignant tumor in human beings. To cure of this disease, we developed an orally absorbable heparin that could show antiangiogenic activity. Firstly, lactoferrin (Lf) was conjugated with heparin that is known as an anti-angiogenic molecule because of its binding property to the growth factors. Lf can be orally absorbed by lactoferrin receptor that is expressed on the intestine, blood-brain barrier and GBM. Using these properties, Lf-heparin conjugate delivered to the GBM by receptor-mediated transcytosis. Through this study, we confirmed conjugation between Lf-heparin as amide bonds and this material can inhibit angiogenesis effectively without toxicity. Also, Lf-heparin could attenuate angiogenesis progression in the GBM orthotopic mouse model. Collectively, these results demonstrated the Lf-heparin would be a novel remedy for the successful outcome of brain cancer. |
