Small interfering RNAs (siRNAs) have been used to selectively regulate the gene expression by a sequence specific mRNA cleavage. To this end, a variety of viral vector and non-viral vector such as adenovirus, retrovirus, cationic polymers, lipids have been investigated for efficient intracellular delivery of siRNAs into the cells. However, the current delivery systems had multiple unwanted problems of cellular toxicity, immune response, and non-specificity towards tissue and cells. In this study, we introduced carbon based materials (Graphene Quantum Dot(GQD) and Graphene Oxide(GO)) on siRNAs. The conjugated materials (siRNA-GQD or siRNA-GO) show hyperthermal property in which might facilitate the endosomal release of siRNA with negligible cytotoxicity. Effective gene silencing under a NIR laser exposure has been investigated in detail. |
