Nitric oxide (NO) has emerged as an important signaling molecule for the control of tumor progression. High concentrations of NO can mediate the inhibition of tumor growth and cancer cell apoptosis. In this work, we selected GSNO as an endogenous NO donor. GSNO-loaded mineralized nanoparticles were prepared by sequentially adding the aqueous solutions of GSNO and CO32- ions to an aqueous mixture of the block copolymers and Ca2+ ions. The CaCO3 phase of the nanoparticle effectively enveloped GSNO molecules at physiological pH. In contrast, at an endosomal acidic pH, the GSNO release was facilitated due to the rapid dissolution of the CaCO3 phase. The released GSNO from mineralized nanoparticles can be reacted with reducing agents, such as glutathione and ascorbic acid, to generate NO at the cytoplasm. |
