제목 Optical biosensor for single mismatched breast cancer type 1 gene detection
초록 Hybridization events on gold nanoparticles (AuNPs) modified with single strand DNA (ssDNA) are simply confirmed on a well-designed glass sensor since the density of AuNPs can be numerically determined based on surface plasmon resonance. In this nanoscale platform, we can identify hybridization between target ssDNA and detection probe in a fixed orientation. Because the hybridization efficiency remarkably depends on the electrostatic interaction, we can easily change it by adjusting the salt concentration of incubation solution. Due to the stability differences between single mismatched DNA (SMT) and perfectly matched DNA (PMT), the hybridization efficiencies are also different. Thus, we further investigated the hybridization efficiencies in the presence of different buffers, and obtained the optimized salt concentration allowed for discrimination of SMT from PMT. As a result, we can apply this strategy to diagnose genetic diseases on AuNP- based biosensors.
