제목 Thermally cross-linked superparamagnetic iron oxide nanoparticles (TCL-SPION) as a dual cancer imaging nanoprobe
초록 A multimodal nanoprobe that can detect cancers by both magnetic resonance (MR) imaging and optical fluorescence imaging is presented in this report. A new anti-biofouling polymer, poly(TMSMA-r-PEGMA-r-NHS) synthesized by radical polymerization was used to coat magnetite (Fe3O4) SPION and subsequently, the resulting polymer-coated SPION was further heated above 80℃ to make the final TCL-SPION through cross-linking between silane groups in the polymer coat. Carboxyl groups of TCL-SPION were converted to amine functionalized TCL-SPION which subsequently reacts with near IR dye, Cy5.5 to generate Cy5.5 conjugated TCL-SPION. After i.v. injection of the nanoprobe to LLC bearing nude mice, tumor site was preferentially detected in IVIS optical image as well as in T2-weighted MR image, showing ~68 % signal. Therefore, we expect that the exploitation of such a stable and effective dual imaging nanoprobe would provide great potential for early cancer diagnosis.
