Effect of Preparation Condition on Release Behavior of Tramadol HCl-loaded Polyoxalate Microspheres |

Tramadol HCl(TH)-loaded Polyoxalate(POX) microspheres were prepared by W1/O/W2, O1/O2/W, O/W and O1/O2 solvent evaporation method for sustained release. We investigated the release behavior according to POX molecular weight and preparation condition. TH-load POX microspheres by O1/O2 solvent evaporation method had smooth surfaces and double layered structure. The microsphere was characterized on the surface, the cross-section morphology, the behavior of TH release for 2 weeks by SEM and HPLC. As the POX molecular weight increased, the release rate of TH decreased. The behaviors of degradation were decreased according to increased POX molecular weight. The analysis of crystallization was analyzed to use X-ray diffraction, differential scanning calorimeter and fourier transform infrared spectrometer. In this study, the results showed that the release behaviors can be controlled by various of molecular weight and concentration of POX. |
