제목 | Polyethylenimine Derivatives as Vectors for Gene Delivery |
초록 | Cationic polymers, such as PEI, are receiving the growing attention as vectors for gene therapy. PEI as cationic source complexes with anionic phosphate of DNA through electrostatic interaction, resulting in a reduced size of the DNA and an enhanced stability toward nucleases. Efficient delivery of foreign DNA into cells is an extremely critical step in gene therapy. To avoid using viral vector systems that may elicit undersirable safety problems, non-viral vector systems are an attractive alternative method to deliver DNA into cells. DNA condensation of PEI was slightly impaired at lower N/P ratio. We measured the complex size and zeta-potential of polymer-DNA complexes. We also investigated the cytotoxicity tested by the MTT method and the transfection efficiency in rat bone marrow stromal cells (BMSCs), muscle-drived stem cells (MDSCs) and NIH/3T3 fibroblast cell line. The transfection studies showed that the polymer-DNA complexes are capable of transfecting for cells used. PEI proved to be an efficient gene delivery system in various cells.
