Preparation of poly(ethylenimine) modified by methoxy-poly(ethyleneglycol) and folic acid as a gene delivery carrier. |

Recently the gene therapy using non-viral vectors draws many attentions due to the unsafety problems of viral vectors. Among the non-viral vectors, poly(ethylenimine) (PEI) is considered to be a good candidate for the gene delivery system. In this study, PEI modified by methoxy-poly(ethyleneglycol) (MPEG) and folic acid as gene carrier was synthesised to decrease a toxicity for cell and to improve in vivo targeting. First, MPEG was modified by glutaric anhydride (GA) to endow carboxylic end group, followed by the activation reaction with EDC (N-ehtyl-N'-(3-dimethylaminopropyl) carbodiimide) and NHS (N-hydroxysuccinimide). The activated carboxylic end group of PEG was reacted by the amines of PEI to give PEI modified with PEG (P1). The modified PEI (G1) was synthesized by the reaction of P1 with folate activated through EDC/NHS. The obtained P1 and G1 were characterized by 1H-NMR and FT-IR. The P1 and G1 formed the complex with DNA in various N/P charge ratio. In conclusion, we confirmed that G1 have the possibility as DNA carrier. |
