To application chitosan based gene carrier, chitosan oligosaccharides (COSs) were chemically modified with hydrophobic moiety of deoxycholic acid. The physicochemical properties of the hydrophobized COSs (COSDs) were investigated by using dynamic light scattering and fluorescence spectroscopy. Then, feasibility for biomedical applications were investigated by introducing the COSDs gene delivery system. COSDs formed nano-sized particles in aqueous milieu. The MW of COS and DS were played a critical roles for the physicochemical characteristics of hydrophobized chitosan nanoparticles. The cac values decreased by increasing COSD hydrophobicity. The COSDs efficiently condensed DNA compared to the unmodified COSs. Furthermore, COSDs protected the plasmid DNA form DNase I attack. The cytotoxicity were strongly related with COSD concentration and the MW of COS. Therefore, deoxycholic acid-modified chitosan oligosaccharide can be considered for efficient gene condensing materials for gene delivery systems. |
