Preparation of dexamethasone-loaded PCLA nanofiber sheets for sustained drug delivery |

Recently, electrospinning technology has been widely used as a processing method to make nanofiber sheet (NS) for biomedical application because of its unique features, such as mimicking the structural properties of the extracellular matrix and easy fabrication process. For these reasons, we prepared the controllable biodegradable poly(ε-caprolactone-co-L-lactide) (PCLA) copolymer. To develop a sustained dexamethasone (Dex) delivery system in this work, we prepared electrospun Dex-loaded NS using PCLA copolymer and water insoluble Dex (Dex(b)) or soluble Dex (Dex(s)). In the evaluation Dex release from Dex-NS, Dex(b)-NS showed the initial burst of Dex(b) at 1 day and maintained almost same amounts of Dex(b) for up to 28 days. On the contrary, Dex(s)-NS exhibited little initial burst of Dex(s) and first order releasing profile from Dex-NS. In conclusion, Dex-NS exhibited the sustained Dex(s) releasing for prolonged action periods as well as controlled biodegradation of the NS. |
