제목 PHA synthase 활성에 의한 polyhydroxybutyrate (PHB)의 분자량 및 분자량 분포의 in vivo 제어
초록 A synthetic operon for polyhydroxyalkanoate (PHA) biosynthesis designed to yield highlevels of in vivo PHA synthase activity was constructed by positioning a genetic fragmentencoding β-ketothiolase and acetylCoA reductase behind a modified synthase gene containingan Escherichia coli promoter and ribosome binding site. Plasmids containing the syntheticoperon and the native Alcaligenes eutrophus PHA operon were transformed into E. coli DH5αand analyzed for polyhydroxybutyrate production. The molecular weight of polymer isolatedfrom recombinant E. coli containing the modified synthase construct was lower than that ofthe polymer from E. coli containing the native A. eutrophus operon. Comparison of theenzyme activity levels of PHA biosynthetic enzymes in a strain encoding the native operonwith a strain possessing the synthetic operon indicates that the amount ofpolyhydroxybutyrate synthase in a host organism plays a key role in controlling the molecularweight of polymer.
