Polypeptide-based amphiphilic copolymer as a pH-sensitive drug carrier |

Polymeric micelles have been investigated extensively in the past years. Smart polymeric nanocarriers which are responsive to certain external stimuli are more efficient to deliver and release the encapsulated drugs to the pathologic area. We successfully synthesized a series of amphiphilic pH-sensitive diblock copolymer based on poly(L-glutamic acid). These block polymers were characterized by 1H NMR and GPC. The substitution ratio can be easily controlled by changing the reaction time. Dynamic light scattering (DLS) showed that the sizes of the micelles formed from these polymers were between 40nm to 50nm at pH 7.4 PBS and were responsive to pH. Doxorubicin (DOX) was encapsulated in the micelles. And the release of DOX was much faster at pH 6.4 than pH 7.4 which made them good candidates as a drug carriers. |
