제목 M13 Bacteriophage/ Silver Nanowire Surface Enhanced Raman Scattering (SERS) Biosensor for Sensitive and Selective Pesticide Detection
초록 M13 bacteriophage-functionalized silver nanowire SERS sensor was fabricated by simply mixing and vacuum filtration of silver nanowire (AgNW) and M13 bacteriophage solution. Tryptophan – histidine - tryptophan (WHW) peptide sequence expressing M13 bacteriophage (BPWHW) was prepared by genetic engineering of surface protein and it functioned as bio-receptor on BPWHW-functionalized AgNW (BPWHW/AgNW) SERS substrate. The prepared BPWHW/AgNW substrate was characterized by scanning electron microscopy (SEM), confocal scanning fluorescence microscopy (CSFM), atomic force microscopy (AFM), and fourier transform infrared (FT-IR) spectroscopy. The BPWHW/AgNW exhibited significantly enhanced Raman signal by the formation of high density hot spots at the AgNW intersections. Limit of detection (LOD) of Rhodamine 6G (R6G) and paraquat (PQ) pesticide on BPWHW/AgNW was 10 nM and 1 ppm, respectively, and exhibited linear relation depending on the R6G and PQ concentrations. Simultaneously, BPWHW/AgNW could selectively capture the PQ through the BPWHW receptor whereas wild-type M13 bacteriophage-functionalized AgNW (BPWT/AgNW) and control AgNW showed no significant binding affinity with PQ. Finally, SERS signal of PQ on the apple peels was detected by wiping with water wetted BPWHW/AgNW substrate. The detection limit of PQ transferred from apple peel was 0.1 μg/cm2, which was much smaller than the maximum residual limit (MRL) of PQ in USA, EU, and China. In conclusion, the developed biosensor is expected to be widely applied for various pesticides and chemicals detection sensor by changing peptide sequence of M13 bacteriophage.
