Stable drug nanoparticles can be conveniently prepared by nano-comminution in the presence of polymeric stabilizers. In this study, the nano-comminution method was further developed to prepare polymeric empty cages and surface modified drug nanoparticles. Drug nanoparticles were first prepared to have polymer chains adsorbed on their surface. Then, the absorbed polymer was crosslinked by ionic gelation with tripolyphosphate anions(TPP). Empty cages were prepared by removing drug nanoparticle cores within the crosslinked polymers. Empty cages can be loaded with different materials. In another approach, folic acid as a ligand for targeting cell membrane was conjugated with the polymer. Conjugation of polymer and folic acid was checked by 1H-NMR. These functional drug nanoparticles can be transported specifically to tumor cells. |
