Multi-arm polyethylene glycol-linear polyethyleneimines(MAPEG-LPEIs) with water-soluble, high transfection efficiency and low cytotoxicity were designed and synthesized as a nonviral gene delivery carriers. The cationic polymers were prepared by coupling six-arm PEG-NHS(10K) to the amino groups of linear PEI(2.5K) with controlled composition. Two copolymers, MAPEG-LPEI3 and MAPEG-LPEI6, were synthesized and their molecular weights were 17.5K and 25K relatively . The polyplex from MAPEG-LPEI3, MAPEG-LPEI6 and plasmid DNA (pDNA) stably dispersed in aqueous medium with narrowly distributed size range of ~200 nm as determined by dynamic light scattering. The polyplexes were able to efficiently transfect luciferase-coded reporter gene into Hela cancer cells in vitro. MAPEG-LPEI6 showed higher transfection activity than MAPEG-LPEI3 at same w/w ratios. Furthermore, MAPEG-LPEIx/pDNA polyplexs were less toxic than LPEI/pDNA complexes as determined by MTT assay. |
