제목 Controllable and Localized Co-Delivery of TRAIL and 6-MP Using Coacervate-Embedded Composite Hydrogels for Enhanced Anticancer Therapy
초록 Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) selectively induces apoptosis of cancer cells. However, a short half-life and low in vivo stability of TRAIL limit its functional therapeutic outcomes. To overcome these drawbacks, we designed a TRAIL-loaded coacervate(Coa)-embedded hydrogel platform for localization and sustained delivery of anticancer drugs. Herein, we used (1) Coa consisting of a arginine-based cationic polymer and heparin for encapsulating TRAILs and (2) thiolated gelatin/poly(ethylene glycol) diacrylate interpenetrating composite hydrogels with anti-cancer chemo-drug 6-mercaptopurine (6-MP) for an injectable reservoir. This delivery system could effectively maintain bioactivity of exogenously delivered TRAILs and control the optimal therapeutic dose in the tumor microenvironment. In summary, our injectable hydrogel platform using Coa-mediated exogenous TRAIL delivery and 6-MP could be a synergistic application for efficient anti-cancer therapy.
