Chitosan, a natural polymer, is a non-toxic, biodegradable polysaccharide with low immunogenicity. Chitosan is also considered to be a good candidate for gene delivery system, since cationically charged chitosan can be complexed with negatively charged plasmid DNA. In this work, artery wall cells targeted gene carrier was synthesized to therapy the disease related with cardiovascular, such as atherosclerosis, restenosis using low molecular, water-soluble chitosa (LMWSC). A molecular weight (Mw) and degree of deacetylation (DDA) of LMWSC were measured by relative viscometer and Kina titration. The structure of LMWSC was analyzed by measuring FT-IR, 1H-NMR, and 13C-NMR. AWBP-PEG-g-LMWSC was synthesized by conjugation the artery wall binding peptide (AWBP), a specific targeting peptide, to the end of PEG-g-LMWSC as a gene carrier to target artery wall cells. Characters of the synthesized AWBP-PEG-g-LMWSC were analyzed by measuring FT-IR, 1H-NMR, zeta-potentiometer, and AFM. The data from this study provided useful and important information for the future application of in vitro, in vivo studies.
