제목 Improving Gene Silencing Efficiency of PEGylated siRNA Complexed with Temperature-Sensitive Pluronic/Poly(ethylenimine) Nanocapsules: Cold Shock Induced Endosome Breaking Effect
초록 Pluronic/PEI nanocapsules (NCs) containing a hollow interior structure were synthesized by cross-linking between pre-activated Pluronic F-127 and poly(ethylenimine) at an oil-in-water interface during a modified emulsification/solvent evaporation process. These NCs exhibited reversible swelling/deswelling volume transition in response to temperature. The size of NCs measured by DLS was about 160 nm at 37 °C and about 400 nm at 15 °C. This transition was caused by temperature-dependent hydrophobic interaction of Pluronic grafted to NC shell layer. We previously proved that Pluronic/PEI NCs could be utilized as an endosome breaking agent by lowering the temperature below a phase transition temperature, which triggers rapid volume expansion upon cooling. PEGylated VEGF or GFP siRNA (siRNA-PEG) was complexed with Pluronic/PEI NCs, delivered into the cells, and the gene silencing effect was comparatively examined with and without the cold shock treatment to verify the hypothesis.
