제목 Di-/tri-block Copolymer of PEGylated siRNA
초록 Small interfering RNAs (siRNAs) are full of promise for the genomic therapeutics because of their high sequence-specificity and inhibition-efficiency to the target cell in mRNA level. But they could be easily broken in the course of the delivery to the target cell and their anionic characteristics prevent the cellular uptake. In our previous studies, the poly(ethyleneglycol) (PEG) was conjugated to the 3’ end of siRNA for protecting from the RNA degradation by the biodegradable-disulfide bond between PEG and siRNA. Now we conjugated PEG to single strand siRNAs via disulfide bond and synthesized A-B and A-B-A copolymer of PEGylated siRNA by hybridization. Then we compared gene silencing effects and serum stabilities between naked and di-/tri-block copolymer of PEGylated siRNA.
